RESUMO

Capillary electrophoresis has been used to directly identify and measure the neurotransmitter, dopamine, in two vesicular compartments in a single nerve cell of Planorbis corneus. Dopamine in the cytoplasm and in easily released transmitter vesicles was separated from dopamine in what are apparently non-functional storage vesicles. In this method, the two peaks in the electropherogram attributed to dopamine were differentiated based on cell lyse time in a non-physiological buffer. The measurements presented here suggest that of the total dopamine present in the cell: 24% is in the cytoplasmic and easily released compartments and 76% is more centrally located, perhaps in a reserve compartment, in the cell. This methodology provides the means to determine molecular species in subcellular compartments and should allow kinetic parameters associated with membrane lysing to be evaluated at single nerve cells.

Biopsy specimens from nasopharyngeal carcinomas (NPC) or salivary-gland carcinomas (SGC) in Greenland Eskimoes were examined for the presence of Epstein-Barr virus (EBV) DNA and sera from the patients were tested for EBV-specific antibody titres. Six out of 7 NPCs and one from an undifferentiated SGG were positive for EBV DNA. The EBV-specific antibody spectra and titres of the patients with NPC or undifferentiated SGG conformed to the results of earlier studies in other high-incidence areas.

RESUMO

The results of operations for congenital deformities of the external and middle ear throughout a 10-year period, 1962-1972, are presented. 38 ears were operated upon, of which 26 obtained an average hearing gain of 25 dB. In 12 ears no improvement was obtained, and no ear became worse. Among the 15 patients with bilateral deformities 11 obtained a postoperative hearing in one or both ears of 40 dB or better, in average 30 dB.

As a continuation of our previous work, where we have demonstrated that in chronic otitis media the picture in the submucosa-bone marginal zone is dominated by capillary proliferation and occurrence of a mononuclear, histiocyte-like cell containing lysosome-like cytoplasmatic bodies, we now report the presence of considerable activity of acid phosphatase in close relation to the eroded bone. The activity was localized both extracellularly, spread along the bony surface, as well as intracellularly in mononuclear, histiocyte-like cells. The acid phosphatase is the "marker" enzyme for lysosomes, and cells with these lysosomes guarantee the presence of enzymatic activity capable of attacking bone collagen. It is difficult to avoid the conclusion that the lysosomes and their enzymes are directly involved in the processes of bone resorption.
